Protect your Ends! The Role of Human Oncofetal HMGA2 in Cancer Genome Stability and as Cancer Drug Target 
 
Abstract: High-mobility group AT-hook (HMGA) proteins are evolutionarily highly conserved in mammals and normally expressed in pluripotent stem cells and during embryonic/fetal stages, when they serve important roles in gene regulation and cell differentiation. HMGA2 is absent from adult somatic tissues, but aberrantly re-expressed in many aggressive human neoplasias due, at least in part, to let-7 miRNA degradation. Furthermore, HMGA2 expression levels causally correlate with the degree of malignancy and metastatic potential. Hence, the protein has recently emerged as cancer drug target. Here, I will describe how HMGA2 serves an important role in human genome stability by effectively protecting difficult-to-replicate chromosomal segments at telomeric regions from replication fork collapse. The fact that HMGA2-positive cancer cells hijack this function to prevent death from induced replication stress, in combination with results from our novel cell-based high-throughput compound screening for HMGA2 antagonists, pave the way for potential high precision targeting of HMGA-expressing malignancies in the context of personalized medicine.
